Stability During Prolonged Incubation
The stability of restriction enzymes in a reaction mixture depends upon the nature of the enzyme, the buffer composition and the incubation temperature.
If a restriction enzyme retains its activity in the reaction mixture for more than one hour, DNA can be digested with less enzyme, by using a prolonged incubation period. For exact quantities of enzymes sufficient for overnight digestion, refer to the table "Reaction Conditions for Restriction Enzymes".
