To prepare sterile growth media, the use of clean glassware is sufficient. Sterile glassware is not required.
Pour the pouch contents into a 500 ml glass bottle or flask.
- Add 200 ml deionized or distilled water.
- Heat the medium in a microwave oven on MEDIUM power setting until bubbles start to appear (2-3 minutes). Swirl gently to mix the preparation.
- Reheat the medium for 30 seconds and swirl gently again. Do not overboil.
- Repeat step 4 if necessary until the medium is completely dissolved.
- Allow the medium to cool and pour plates (for solid media) or use directly (for liquid media).
For solid media, leaving plates in a sterile air flow cabinet with the lids slightly open allows for faster solidification of the media and prevention of water condensation on the plates.
Do not autoclave!
