Description
0.5 M EDTA, pH 8.0, is 0.22 µm membrane-filtered aqueous solution of ethylendiaminetetraacetic acid (EDTA), sodium salt. It is suitable for biochemistry or molecular biology applications that require a chelator for divalent cations. By forming complexes with metal ions, EDTA inhibits metal-dependent enzymatic reactions. The EDTA complexes metal ions at a 1:1 ratio.
Nucleic acids are usually stored in TE buffer that contains 1 mM EDTA.

Quality Control
The absence of endodeoxyribonucleases, exodeoxyribonucleases and ribonucleases is confirmed by appropriate quality tests.

Storage
Store at +4°C.

Usage Recommendations
For immediate inhibition of metal-dependent enzymatic reactions, add 1 µl of 0.5 M EDTA to 20 µl reaction mixture. Mix well.

Note

  • For visualization of the reaction products on an agarose gel:
    • mix 5 volumes of the reaction mixture with 1 volume of the 6X DNA Loading Dye;
    • load the mixture on a gel and run electrophoresis.
  • For usage of the reaction products in further enzymatic reactions:
    • after the addition of EDTA, the reaction mixture should be purified using Fermentas DNA Gel Extraction Kit or by phenol/chloroform extraction and alcohol precipitation (see protocol).
  • For storage of reaction mixtures:
    • after the addition of EDTA, all enzymes in the reaction mixture should be thermally inactivated if possible.
