RiboRuler™ RNA Ladder, Low Range
The RiboRuler™ Low Range RNA Ladder is a mixture of seven chromatography-purified single-stranded RNA transcripts. The RNA concentration of each ladder band is precisely measured by spectrophotometry (see photos below), which makes this ladder ideal for approximate RNA quantification on gels.
The RiboRuler™ Low Range RNA Ladder is recommended for electrophoresis in the following: native 2% agarose with TAE buffer, denaturing formaldehyde agarose with MOPS buffer, denaturing glyoxal/DMSO agarose with sodium phosphate buffer and denaturing polyacrylamide gel electrophoresis in TBE buffer.
The ladder is supplied with 2X RNA Loading Dye.

The RiboRuler™ High Range RNA Ladder is a mixture of eight chromatography-purified single-stranded RNA transcripts. The RNA concentration of each ladder band is precisely measured by spectrophotometry (see photos below), which makes this ladder ideal for approximate RNA quantification on gels.
The RiboRuler™ High Range RNA Ladder is recommended for electrophoresis in the following: native 1% agarose with TAE buffer, denaturing formaldehyde agarose with MOPS buffer and denaturing glyoxal/DMSO agarose with sodium phosphate buffer.

Description
RiboRuler™ RNA ladders are mixtures of chromatography purified single-stranded RNA transcripts, produced from specific templates that contain a fragment of the pTZ19R polylinker and lambda phage fragments. These ladders are ideal for qualitative and quantitave analysis of RNA both in native or denaturing agarose and polyacrylamide gels. An optimized sequence guarantees the migration of the ladder bands according to their actual size in a variety of electrophoretic conditions. Chromatography purified RiboRuler™ RNA ladders are free of degraded RNA and NTPs; the RNA concentration of each ladder band is precisely measured by spectrophotometry, making the RiboRuler™ RNA ladders ideal for approximate RNA quantification on gels.
All RNA ladders are supplied with 2X RNA Loading Dye. The inclusion of ethidium bromide in the 2X RNA Loading Dye allows visualization of the RNA without additional staining of denaturing gels. For better RNA visualization on native gels, additional staining with ethidium bromide is recommended.
Ready-to-use RiboRuler™ RNA ladders are premixed with RNA Loading Dye. After denaturation for 10 min at 70°C and chilling on ice, ready-to-use RiboRuler™ RNA ladders can be directly applied onto the gel (see Protocols for RNA Electrophoresis).
RiboRuler™ RNA ladders (except the ready-to-use versions) are suitable for end-labeling using T4 Polynucleotide Kinase (see protocol) making them ideal for Northern blots.
All RiboRuler™ RNA ladders are aliquoted in smaller volumes to minimize the possibility of contamination during usage.Storage Buffer (for conventional RNA ladders)

47.5% formamide, 0.0125% SDS, 0.0125% bromophenol blue, 0.0125% xylene cyanol FF, 0.0125% ethidium bromide and 0.75 mM EDTA.2X RNA Loading Dye
95% formamide, 0.025% SDS, 0.025% bromophenol blue, 0.025% xylene cyanol FF, 0.025% ethidium bromide and 0.5 mM EDTA.Recommendations for Loading (see Protocols for RNA Electrophoresis)
- Use the supplied 2X RNA Loading Dye for sample RNA and ladder RNA. Loading equal volumes of sample RNA and ladder RNA is recommended.
- For RiboRuler™ RNA Ladders:
Use 0.25 µl of RNA ladder per 1 mm of gel lane width.
- Mix 2 µl of RNA ladder and 2 µl of the 2X RNA Loading Dye.
- Heat RNA ladders at 70°C for 10 min.
- Chill on ice and centrifuge briefly prior to loading onto the gel.
- For ready-to-use RiboRuler™ RNA Ladders:
Use 0.5 µl of ready-to-use RNA ladder per 1 mm of gel lane width.
- Heat RNA ladders at 70°C for 10 min.
- Chill on ice and centrifuge briefly prior to loading onto the gel.
Quality Control
RiboRuler™ Low Range RNA Ladders are tested on native agarose and polyacrylamide gels.
RiboRuler™ High Range RNA Ladders are tested on native agarose gels.
RNA concentration is determined spectrophotometrically. The absence of ribonucleases is confirmed by appropriate tests.Storage and Stability
Store at -20°C for 6 months, or at -70°C for 24 months.
Ready-to-use RiboRuler™ RNA ladders, premixed with 2X RNA Loading Dye, are stable at +4°C for approximately 2 months.
Note
Any RNA, including RiboRuler™ RNA ladders, is extremely sensitive to degradation by ribonucleases. The use of fresh electrophoresis buffer, freshly poured gels, diethyl pyrocarbonate (DEPC)-treated solutions and protective gloves minimizes the possibility of contamination. Gloves are also necessary when solutions containing ethidium bromide are handled.
