FastRuler™ DNA Ladder, Ultra Low Range, ready-to-use

Designed for fast sizing and approximate quantification of double-stranded DNA in 48-96-well high throughput gels and in conventional agarose gels.
DNA marker contains dephosphorylated DNA fragments and is ideal for forward 5'-end labeling with T4 Polynucleotide Kinase (see protocol below).
FastRuler™ Ultra Low Range DNA Ladder, ready-to-use, is supplied with 6X Orange DNA Loading Dye.

Protocol for Forward Radioactive 5'-termini Labeling using T4 Polynucleotide Kinase

  1. Prepare the following reaction mixture:
    FastRuler™ Ultra Low Range DNA Ladder, ready-to-use 2 µl
    10X Reaction Buffer A (supplied with the enzyme) 2 µl
    [gama-32P]-ATP or [gama-33P]-ATP 2 µl (3.3 pmol/µl)
    Water, nuclease-free 13 µl
    T4 Polynucleotide Kinase 1 µl (10 u)
  2. Incubate at 37°C for 30 minutes.
  3. Add 1 µl 0.5 M EDTA, pH 8.0, and extract with an equal volume of chloroform.
  4. Purify the labeled DNA on Sephadex™ G-50 column.

Description
FastRuler™ DNA ladders are specifically designed for fast sizing and approximate quantification of double-stranded DNA in 48-well (or 96-well) high throughput gels, as well as in conventional agarose gels. The ladders are mixtures of five blunt-ended chromatography-purified individual DNA fragments that are easily identified after the 8-14 min run on the appropriate agarose gels in a short separation distance (10-20 mm). Supplied with the 6X MassRuler™ DNA Loading Dye or with the 6X Orange DNA Loading Dye for the sample DNA.
FastRuler™ Ultra Low Range DNA Ladder, ready-to-use, contains dephosphorylated DNA fragments and is ideal for forward 5'-end labeling with T4 Polynucleotide Kinase (see protocol).

Storage and Loading Buffer
10 mM Tris-HCl (pH 7.6), 10 mM EDTA, 0.005% bromophenol blue, 10% glycerol.

6X MassRuler™ DNA Loading Dye
10 mM Tris-HCl (pH 7.6), 0.03% bromophenol blue, 60% glycerol and 60 mM EDTA.

Storage and Loading Buffer (for FastRuler™ Ultra Low Range)
10 mM Tris-HCl (pH 7.6), 10 mM EDTA, 0.025% orange G, 0.005% xylene cyanol FF, 10% glycerol.

6X Orange DNA Loading Dye
10 mM Tris-HCl (pH 7.6), 0.15% orange G, 0.03% xylene cyanol FF, 60% glycerol and 60 mM EDTA.

Recommendations for Loading

Quality Control
Tested in appropriate gel electrophoresis. The DNA concentration is determined spectrophotometrically. The absence of nucleases is confirmed by appropriate tests.

Storage
Store at room temperature or at +4°C for 6 months. For longer periods, store at -20°C.
