Electrophoresis of tracking dyes in 6X Orange DNA Loading DyeDescription
The 6X Orange DNA Loading Dye is used to prepare DNA markers and samples for loading on agarose or polyacrylamide gels. It is recommended for the analysis of small DNA molecules. The solution contains two dyes, orange G and xylene cyanol FF for visual tracking of DNA migration during electrophoresis. The presence of glycerol in the solution ensures that the same sample forms a layer at the bottom of the well; the EDTA included in the solution binds divalent metal ions and inhibits metal dependent nucleases.Composition of 6X Solution
10 mM Tris-HCl (pH 7.6), 0.15% orange G, 0.03% xylene cyanol FF, 60% glycerol, 60 mM EDTA.
Note
- In 1% agarose gels orange G co-migrates with ~50 bp DNA, while xylene cyanol FF co-migrates with ~4000 bp DNA.

Quality Control
Tested for DNA sample preparation prior to agarose gel electrophoresis.
Storage
Store at room temperature +4°C up to 12 months.
For longer periods, store at -20°C.
Usage Recommendations
- Add 1 volume of 6X Orange DNA Loading Dye to 5 volumes of DNA sample.
- Mix well, spin down and load.
