Electrophoresis of tracking dyes in 6X MassRuler™ DNA Loading Dye

Description
The 6X MassRuler™ DNA Loading Dye is used to prepare DNA markers and samples for loading on agarose or polyacrylamide gels. It is recommended for DNA quantification. The solution contains a single dye, bromophenol blue, that minimizes possible DNA fragment’s masking under UV. The presence of glycerol in the solution ensures that the same sample forms a layer at the bottom of the well; the EDTA included in the solution binds divalent metal ions and inhibits metal dependent nucleases.

Composition of 6X Solution

10 mM Tris-HCl (pH 7.6), 0.03% bromophenol blue, 60% glycerol, 60 mM EDTA.

Note

• For precise quantification it is recommended to load equal volumes of sample DNA and marker/ladder DNA.
• If necessary, adjust the concentration of your sample so that the expected amount of DNA loaded is approximately equal to that of a DNA ladder band of the nearest size.
• In 1% agarose gels bromophenol blue co-migrates with ~300 bp DNA.

Storage
Store at room temperature +4°C up to 12 months.
For longer periods, store at -20°C.

Quality Control
Tested for DNA sample preparation prior to agarose gel electrophoresis.

Usage Recommendations

Add 1 volume of 6X MassRuler™ DNA Loading Dye to 5 volumes of DNA sample. Mix well, spin down and load.
